Pros
No-nonsense, elegant, customizable, works great with pop3/imap, A BLESSED RELIEF from the stupidity of Windows Mail and other insufferable, cutsey programs.
Also the aesthetics are great -- elegant, clean, neat.
Cons
In my use, I've come across none.
Summary
I used Outlook Express for years. Once I finally had to give up XP, naturally I had to find a replacement. HATED (!!!) Windows Live Mail. So I tried Thunderbird, which would have been okay except that there's no Outbox -- stuff gets sent immediately, with no "send later" option -- which is a fatal flaw for me. All the other programs had one fatal flaw or another. Finally tried Sylpheed, which fixed everything. VERY grateful for the program.

Note -- regarding Thunderbird, although it is a good program, I much prefer Sylpheed even aside from the Outbox issue.

Pros
Loaded quick, ran fine no bugs, nice interface (I can't remember if it has a spam filter :| ) its opensource so it will always be free, can't beat that. simple setup too. If you set up an E-mail for Thunderbird I just start thinking "HUH???" where do I type my password?, and thunderbird loads slowly.
Cons
no real cons not super spiffy looking but other than that two thumbs up!
